He's been outdone by Great Sable Stag. I can imagine hims being good before m10 though.

First of all, Great Sable Stag is a rare, Karplusan Strider is uncommon, people need to stop declaring cards as worse than similar cards of higher rarity, of course the rarer card is better, that's why it's rarer.
Second, 4 toughness is very significant. Stag is vulnerable to Lightning Bolt, Strider lives through it.
Third, when a Stag and a Strider meet, only the Strider walks away.

For someone playing most of my Magic before the "stack" was introduced: Will the non-targetability be active while the spell is on the stack, or is it an ability of the creature that only takes effect once it enters play?

A decent creature, but obviously can still be countered. In a lot of formats, including Coldsnap's awful Limited, the 4 toughness is quite relevant, and few things could take it out. Furthermore, it was protected from stuff like Chill to the Bone, and blue bounce spells. Not amazing in constructed, but deceptively hard to deal with for certain opponents.
