Can we activate its second ability more than once? Say for example, can I spend 6 colorless mana and have the guardian block 4 total creatures?

Sure, there's no clause preventing you from activating the ability as many times as you have the mana for. The "additional creatures" do stack; it's not worded "Luminous Guardian can block two creatures this turn", or something similar.

@Enemy_Tricolor: Heart of Light is a pretty nice combo. He won't kill anything, but with the recent battles against wither and infect, he's thanked me many times for giving him this.

I'd rather play Palace Guard and stick an Indestructibility on him. Or Righteousness.
